1st thing to do is turn off the "IFC" intelligent frame creation feature in the menu. It's designed for 24fps Blu Ray's but is turned on by default on all your inputs.
Best way to set it up is to use the THX optimiser found on any dvd with a THX logo, like star wars for example. Basically a series of test cards which help you properly tweak all the picture settings. Worked wonders for the picture quality on my Pany. Also try the BBE VIVA 3D virtual surround mode in the sound settings, sounds like stereo but with added surround soundiness. The other fake surround mode sounds terrible.
Other than that the only other thing is you have to do this for all the viewing modes.... normal,cinema,eco etc. I would stay away from the Vibrant mode though. I used "Cinema" mode to set mine up for the 1st time, with the lights off and in the dark which is how I watch movies. Then when I had the settings tweaked using the THX optimiser I used the same settings as a base for "Eco" and "Normal" and tweaked accordingly by eye. Ideally you should use the THX optimiser for all the modes to set them up properly, and make sure you set them up with normal tv viewing light levels in the room.
I use eco mode for watching sky, also handy as it changes the settings depending on the level of light in the room. I use "normal" for playing games on my PS3 as I find the "cinema" settings to dark.
Up to you what you use the different tv modes for, but I find them very handy to use different modes for all my gadgets.
EDIT: Oh and experiment with the picture overscan feature as well, although this only appears with certain tv ratio's. Mainly for normal tv like sky or the digi tuner. You get a better picture with this turned off as the screen isn't zoomed in a bit, but on some programs you might notice jaggly lines right on the edge of the picture. Widescreen programs turn off the overscan feature automatically when it detects the signal, so it only affects some programs. I have mine turned on to remove the jaggily lines on some programs.
Also you can set it to power up on the inbuilt digi tuner or on scart. I don't have an ariel in my tv so I have set this to scart so it starts on sky.
Just work through the manuall it's pretty easy to follow and use the THX optimiser and you will be fine.
And 1 more thing you might need to re tweak your picture settings after the 1st 100 hours or so. Basically the phosphurs in the screen are at the hottest/brightest during the 1st 100 hours or so on a plasma. Mine ran a hell of a lot cooler after 50-100hrs.
