If you mean can you hear more detail in instruments and the studio production quality when encoding to FLAC rather than say MP3, then yes absoluteley.
Any lossless file will sound a lot better than lossy formats, just as 320KBPS sound better than 128KBPS.
With faster internet speeds and bigger hard drives, lossless has to be the way of the future.
Problem is the big labels are still bent on selling us rubbish files over the internet. That'll get me ranting so I'll stop there.
